چکیده

Background Malaria during pregnancy is the cause of 1500 neonatal deaths a day. Moreover, 40% of all low weight births are caused by pregnancy associated malaria. Researchers at Copenhagen University have identified the VAR2CSA protein as a potential protective recombinant placental malaria vaccine. ExpreSion Biotechnologies is responsible establishment of cell lines expressing VAR2CSA variants and for developing the protein production process based on VAR2CSA. The ExpreS System is a one-for-all protein expression system based on Drosophila S2 cells that is excellent in all phases of Drug Discovery, R&D and manufacturing due to high-level transient transfections, easy establishment of stable polyclonal pools that provides continuous high protein expression levels without selection pressure, and simple cloning procedure. It is a novel, nonviral, insect-cell based expression technology applied to the development of a critically needed vaccine. The VAR2CSA protein, which the vaccine is based on, is hard to express and comparison studies between insect, bacteria and yeast have shown that an insect cell system is the only one leading to a clinically useful immune response. Process optimization is also critically important, as the cost of manufacture must be as low as possible to allow the vaccine to be used in the countries where it is most needed.

The Influence of Sub-Unit Composition and Expression System on the Functional Antibody Response in the Development of a VAR2CSA Based Plasmodium falciparum Placental Malaria Vaccine

The disease caused by Plasmodium falciparum (Pf) involves different clinical manifestations that, cumulatively, kill hundreds of thousands every year. Placental malaria (PM) is one such manifestation in which Pf infected erythrocytes (IE) bind to chondroitin sulphate A (CSA) through expression of VAR2CSA, a parasite-derived antigen. Protection against PM is mediated by antibodies that inhibit b...

Expression of S and pre s2 Hepatitis B Surface Antigens in Mammalian Cos-7 Cell Line

Hepatitis B virus (HBV) is a serious global health problem. The development of a safe and effective vaccinewould help infection prevention. Previous hepatitis B vaccine production involved the isolation of the noninfectious particle from chronic HBV carriers. DNA recombinant technology has been used for vaccineproduction without having been contaminated with blood-born infectious ag...

Production of full-length soluble Plasmodium falciparum RH5 protein vaccine using a Drosophila melanogaster Schneider 2 stable cell line system

The Plasmodium falciparum reticulocyte-binding protein homolog 5 (PfRH5) has recently emerged as a leading candidate antigen against the blood-stage human malaria parasite. However it has proved challenging to identify a heterologous expression platform that can produce a soluble protein-based vaccine in a manner compliant with current Good Manufacturing Practice (cGMP). Here we report the prod...

Expression of a New Recombinant Collagenase Protein of Lucilia Sericata in SF9 Insect Cell as a Potential for Wound Healing

Background: Today, the use of maggot therapy has become widespread due to the increase in chronic ulcers in the world. The recombinant production of secreted enzymes from these larvae is a novel non-invasive method for the treatment of chronic ulcers. Lucilia sericata (L. sericata) collagenase (MMP-1) has been expressed in insect cells. Collagenase is an enzyme that is widely used in clinical t...

HIV-1 virus-like particles produced by stably transfected Drosophila S2 cells: a desirable vaccine component.

The development of a successful vaccine against human immunodeficiency virus type 1 (HIV-1) likely requires immunogens that elicit both broadly neutralizing antibodies against envelope spikes and T cell responses that recognize multiple viral proteins. HIV-1 virus-like particles (VLP), because they display authentic envelope spikes on the particle surface, may be developed into such immunogens....
